Localized Reconstruction is a general method for the localized three-dimensional reconstruction of substructures bound to a larger particle. After determining the particle orientations via conventional methods, local areas corresponding to the subunits ('subparticles') can be extracted and treated as single particles.
The method has been incorporated in Scipion version 1.1 and the relevant protocols are available under 'opic'.
If you have any questions regarding the method or its command line use as described here, please don't hesitate to contact us at [email protected].
These instructions are for version 1.2.0 and for running the script from command line.
